The Lady Panthers (13-0, 2-0) out-slugged Nashville 15-9, twice rallying from early inning deficits.
PCHS will host Du Quoin in a battle of River-to-River Conference, Mississippi Division undefeated squads Friday.
Nashville's records are 5-2 and 1-1.
Mo Ramsey was 3-for-4 with four RBI for Pinckneyville with teammates Emily Miller, Sara Malanowski and Dani Jenkel all adding two-hit efforts.
Malanowski-also the winning pitcher in relief of Emily Chapman-knocked in four runs and homered.
"The wind was gusting about 40 MPH," Lady Panthers coach Greg Hale said. "It was just difficult to play today, especially with so many balls hit into the air."
Pinckneyville was behind 3-1 after one inning and 7-6 to start the fourth. In the last four frames, the Lady Panthers touched Nashville pitchers Breanne Pelker and Shaye Harre for nine runs.
"We offensively are very happy with our output today," Hale said. "Anytime you can score 15 runs, good things are going to happen."
Taylor Cicardi, Maddy Keith, Chapman, Macie Minor and Taylor Cheek also hit safely for PCHS.
