Four people and two boats were involved in the accident, which included major injuries to two people, Franklin County Sheriff Don Jones said.
"The boats were in motion at the time of the accident," Jones said.
Rescue workers were searching an area of the lake north of Route 154 just west of Interstate 57, approximately half a mile north of the Rend Lake Golf Course.
Jones said the boats appeared to be in the channel of Gun Creek when the accident occurred. One boat received major damage, he said.
One male and one female received major injuries and were flown to St. Louis-area hospitals.
The fourth individual, a male, "is missing, and a search is under way to attempt to locate that individual," Jones said.
A ground crew also was searching for him, Jones said.
The Rend Lake, Benton Fire Department and Sesser search-and-rescue teams are involved in the search, as are the Franklin County Sheriff's Office, the U.S. Army Corps of Engineers and the Illinois Department of Natural Resources Law Enforcement Division, Jones said.
